Discovery of  Hexarelin

Hexarelin is a derivative of GHRP-6. Mediolanum Farmaceutici in Spain derived it. This new compound was found to be more stable and well tolerated in comparison to GHRP-6. Hexarelin has reached Phase II of clinical testing and the official results for its application in the treatment of Growth hormone deficiency will be officially reported.

Mechanism of Action

Hexarelin is an analog of growth hormone release peptide (GHRP-6). This molecule produces a potent effect in the release of growth hormones. A study was carried out to understand the mechanism of action of hexarelin. According to this study, hexarelin could potentially increase growth hormone levels. This effect was far more potent than what GHRH produced. However, the effect of hexarelin was short-term.

Based on this experiment, three mechanisms were concluded for Hexarelin. It directly acts on the pituitary gland to release growth hormones. Indirectly, it may also be involved in the release of GHRH. Other than this, it also acts on the hypothalamus to release growth hormones working synergistically with GHRH.

Cardioprotective effects

There have been several experiments that compare hexarelin to ghrelin. However, ghrelin has a very short duration of action which limits its application. Hexarelin, on the other hand, is chemically stable. It has several cardioprotective effects, such as cardiac fibrosis, cardiac dysfunction, ischemic heart disease, and atherosclerosis. The effects of hexarelin, in this case, maybe due to its direct link with the cardiac receptors CD36.

Diabetes

Hexarelin is a regulator of peroxisome-activated receptor gamma. This helps in the regulation of adipogenesis and the metabolism of lipids. Administration of hexarelin helps in the reduction of glucose levels. Also, it helps in increasing the beta cell number that produces insulin. Hence, hexarelin is helpful in the management of diabetes as well as other endocrine and metabolic disorders. Researchers have now increased the application of hexarelin to diabetic cardiomyopathy.
