Discovery of Avanafil:

Avanafil was discovered and approved in the year 2012 by the FDA for the treatment of erectile dysfunction. Furthermore, it got approval from the EMA in the year 2013. Avanafil was invented at the Tanabe Seiyaku Co, which is now known as Mitsubishi Tanabe Pharma. In the United States, Avanafil had exclusive rights for distribution by Metuchen Pharmaceuticals.

Mechanism of action:

Avanafil is a drug (PDEF5 inhibitor) that was approved for the treatment of erectile dysfunction by the EMA and FDA. Avanafil helps in the selective inhibition of PDE5 to inhibit the degradation of cGMP (cyclic guanosine monophosphate), which is found in the corpus cavernosum smooth muscles in the penis. The physiological mechanism behind this action is the release of NO (nitric oxide) in the corpus cavernosum during the time of sexual stimulation. Nitric oxide activates the guanylate cyclase enzyme, which increases the cGMP’s levels. As a result of this, smooth muscles relax in the corpus cavernosum by allowing the inflow of blood into the penis. In comparison to other PDE5 inhibitors, Avanafil has faster action onset.
